However, death still lurks around the Kocho sisters, and Kanae soon loses her life while carrying out a mission as a Demon Slayer. After the death of Kanae, Shinobu still presents a cheerful and gentle demeanor, often seen with a smile that belies the rage and grief she harbors within. Unlike her fellow Hashira, Shinobu lacks the physical strength to decapitate demons. Instead, she employs a unique approach; she uses a special katana that acts as a stinger infused with poison derived from wisteria, a substance lethal to demons. She makes up for her inability to behead demons, reflecting her ability to turn a perceived weakness into a formidable strength, a trait that defines her character throughout the series.

During the final battle in the Infinity Castle, Shinobu faces her sister's killer, Doma. As Shinobu approaches the demon, he is just consuming one of her disciples, an act which enrages the hashira. Shinobu tried her best to fight Doma, but the demon already had counterattacks for the hashira's deadly poisons. Instead of the poison working towards weakening Doma, it is easily nullified and even heals the demon. Despite Shinobu's agility and skill, Doma's regeneration abilities and overwhelming strength make him a formidable opponent.
Moreover, Shinobu knows that a direct fight would be futile given Doma's power, so she tries poisoning him slowly. When Shinobu successfully hits him, the plan, however, comes at a significant cost. Shinobu's ribs and collarbone are cut in the process of injecting Doma with a large amount of poison since small doses of her poison are easily nullified. As Shinobu faces the brink of death, she remains resolute, embodying the spirit of a true Hashira.

Before the final battle against Muzan, Shinobu preempted that he could face Doma, and she prepared ahead due to the demon's powerful abilities. Shinobu's grand plan is the Wisteria poison− she infused the poison into her body as the final attempt to kill him fails. After Shinobu took a lethal blow from Doma, the demon finally consumed her. Shinobu died knowing that Doma would also meet his end. In other words, her death underscores the high stakes of the battle against demons, thus highlighting how vengeance can sometimes lead to death and the immense sacrifices required to protect humanity.
